Reversible lysine acetylation controls the activity of the mitochondrial enzyme acetyl-CoA synthetase 2.
We report that human acetyl-CoA synthetase 2 (AceCS2) is a mitochondrial matrix protein. AceCS2 is reversibly acetylated at Lys-642 in the active site of the enzyme. The mitochondrial sirtuin SIRT3 interacts with AceCS2 and deacetylates Lys-642 both in vitro and in vivo. Deacetylation of AceCS2 by SIRT3 activates the acetyl-CoA synthetase activity of AceCS2. This report identifies the first ace...

Long chain acyl-CoA synthetase 1 (ACSL1) contributes 50 to 90% of total ACSL activity in liver, adipose tissue, and heart and appears to direct the use of long chain fatty acids for energy. Although the functional importance of ACSL1 is becoming clear, little is understood about its post-translational regulation. In order to investigate the post-translational modifications of ACSL1 under differ...
متن کاملReversible inactivation by noradrenaline of long-chain fatty acyl-CoA synthetase in rat adipocytes.
Incubation of rat adipocytes with the same range of noradrenaline concentrations that stimulate lipolysis caused a rapid and stable decrease in the activity of fatty acyl-CoA synthetase. Corticotropin, glucagon and dibutyryl cyclic AMP also decreased the activity of the enzyme. The effect of noradrenaline was apparent over a wide range of concentrations for the three substrates of the enzyme. A...
متن کاملHydrogen ion-induced reversible inactivation and dissociation of formyltetrahydrofolate synthetase.
Formyltetrahydrofolate synthetase undergoes inactivation at pH values below 7. Inactivation is accompanied by both the dissociation of the tetramer into monomers and the exposure of aromatic amino acids to the solvent. The rate of inactivation is a function of pH. This rate and the rate of exposure of tryptophan residues to the aqueous environment follow first order kinetics. After prolonged in...
متن کاملEffect of reversible inactivation of locus ceruleus on naloxone-induced withdrawal syndrome in paragigantocellular neurons in morphine-dependent rats
In this study, the effect of reversible inactivation of locus ceruleus (LC) on naloxone- induced withdrawal syndrome in paragigantocellular (PGi) neurons in morphine- dependent rats was investigated. For inactivation of LC, 1 µl of lidocaine (2%) was used and for induction of withdrawal syndrome, naloxone hydrochloride (2 mg/kg) was injected systemically. The results showed that in dependent gr...
